I guess it doesn't really qualify as a "build", but I have it as one of the environments in my virtual environment room. IM me if you want to see it.

I would love to see something organic like a replica of Mammoth Cave. It's huge cave system in Kentucky, I couldn't really find any pictures that do it justice, but think of the fun to be had with stalagmites and creative lighting effects!

In no particular order:

Pompeii before the eruption
Harry Potter's Hogwarth
2001's space station
The Nostromo from the first Alien movie
Mos Edley's Cantina
The hotel from "The Shining"
The decadent bar from "Clockwork orange" (with AVIs to match :) )
Hopper's Nighthawk's bar

On the wackier side:

A giant, beating, human heart, connected to the land, that we would call "the heart of SL"
The little shop of horror, with an hungry Audrey
Any giant rendition of a still-life from a Flemish master
A giant (Sim sized :) ) computer with very detailed insides reproducing the motherboard, hard disc, etc... It would be great if the details were from an original vintage model, say a Mac 512 or an IBM PC XT.
A giant Concert Piano, again with detailed inwards
A giant woman's purse
A giant sized, detailed Staunton chess set (including knights :D )

Pompeii you say?

Thanks to the fantastic talents of Eva Virgo and Shari Zuma, Koshka and I now have our dream home in Indigo based on an intepretation of historical Pompei mid-eruption complete with ruined columns and a lava flow. The wall art and many of the textures were taken directly from archeological photographs. I hear that ash is good for your complexion.

I have seen a large gorilla, but what about the big walled-in compound as seen in the original King Kong?

Has anyone built Nemo's sub from the sixties version of 20,000 leagues under the sea?

Graceland? The little house on the prarie?

Hmmm, I wonder how prims it would require to do Superman's crystal caverns?

I'm warming up now -- no shame --- recalling the bumper stickers that said "See Rock City". I'll bet this would bring to mind different images depending on age and regional background.

Accually we have be comissioned to build the "Bates Motel".. well at least his "mothers" house. It will be ready later this month. ;)

I would love if someone built a four sim system. Where each sim would be a representation of a different time in Hill Vally: 1885,1955,1985 and 2015. I could spend all of my Second Life playin there *L*.

Maybe somebody like Anshe or Profsky could commission artists to do themed houses by period or region, like Moderniste, Classical, etc...
