Say, one builds a room with four walls out of a single hollowed box prim..
Adding some windows to the walls means that the walls must be split up into multiple prims?
Or, is there some niffty trick to adding a window with a single prim and an alpha texture.

The inside of the hollowed box has to be a single texture. But you could make that so it defines the 4 walls that you want, edge to edge. The repeat factor is normally just under 4x, if using a single wall texture. There is a formula in the LSL WIKKI that says how to calculate that.
The outside is 4 seperate textures.
Yes, it is possible to get different textures on all 4 interior walls this way, and to get the exterior and interior textures to line up. It's also a royal pain to get the adjustments and textures just right.

Since you're probably attempting to reduce prim usage here, you can also consider building the room out of 2 prims that are cut and hollowed to form a C shape. Interlocking them creates a closed box, which you can cut further to reveal tall windows or entrances into the room.
