Is it possible to set the actual MIME type that SL should be expecting when it comes to the Media URL, or are we limited to the Media Type options? I keep getting video/* when I'm putting in some media URLs and they don't work, they sometimes only seem to work when you get something like video/mp4 or the like. Just working with the Media Type doesn't always appear suitable

All media in SL is handled by Quicktime (except Webpages). So if QT can play it, it should be fine. QT is pretty limited, though, if you don't add extra codecs to it (for Mac-Users, the recommended tool is called «Perian» - this plays almost everything).
So, you can't play Windows-Media files, for example, among other stuff...
What are you trying to play, then?
